In the Basic Numbers lesson, sometimes "J'en veux trois..." is used, and other times "Je veux quatre..." is used. Is it a difference in formality, kind of like "I would like..." vs. "I want..."?

In ' On a Tour ' in the interactive section ' J'en suis sur ' means ' I am sure of it ' so I would guess that 'J'en veux trois ' means ' I want three of it ( or them ) ' as opposed to the simpler 'Je veux quatre' 'I want four'

Alan is exactly right. The word 'en' can simply mean 'some' but it also refers to a previously discussed object.
For example, if I were to pose the question, "Tu veux du sucre?" (Do you want some sugar) You could reply, "Oui, j'en veux" (Yes, I want some). As you can see the 'en' functions as a reference to the sugar which is our previously discussed object.
