Why do you put a : in दु:ख?
It seems as if you could just put दुख.

We put a : in दु:ख because दु:ख is a Sanskrit loan word. ः (visarga) has a Ha sound in it.
You transliterate दु:ख as duHkha.
Let look at the word 'Ram" in Hindi versus Sanskrit.
In Hindi we spell Ram as राम.
In Sanskrit we spell Ram as रामः(with ः visarga)
We put a : in दु:ख because दु:ख is a Sanskrit loan word. ः (visarga) has a Ha sound in it.
