If are speaking to an adult stranger or to a superior, you should use usted. Also, anytime you would think of addressing someone using Mr. or Mrs. (or Dr.), you should use usted. When speaking to family members, close friends, or children, you can use informal 'tu' form.

Larryritchie, the lesson specifies you'd be addressing a co-worker, so most likely there is a formal environment. However, it all depends on the closeness you have towards the person you're talking to. If the person is a co-worker and also your friend, you can translate it as "tú".
